    Routes of Dicyandiamide Uptake in Pasture Plants: A Preliminary Laboratory Study

    A consequence of intensification of New Zealand pastures is increased nitrogen (N) inputs to the soil in the form of urine, dung and mineral fertiliser. Dairy cow urine has a high N content that causes large N losses from the grazed system via nitrate (NO3-) leaching, nitrous oxide (N2O) emissions and ammonia volatilization. Dicyandiamide (DCD) is a nitrification inhibitor that has been proven to reduce NO3- leaching and N2O emissions, and increase pasture in New Zealand pastures (De Klein et al., 2014). DCD was commercially available for use in New Zealand pastures until 2013 when its use was suspended due to detection of traces of DCD in exported milk. Although DCD at high doses is relatively non-toxic there is no set maximum residue limit for its consumption. The contamination incident has highlighted the need to understand the pathway by which DCD entered the dairy cow. Nutrients can be absorbed (or taken up) through the leaves via leaf cuticle and stomata of plants (Eichert and Fernández, 2012) and this phenomenon is used to fertilize golf courses and horticultural crops mainly using urea as a spray formulations. Because of the similarity between DCD and urea in terms of molecular weight and structure, we suspected that DCD could similarly be taken up in pasture plants. Few studies have shown the root uptake of DCD but none using pasture plant species. Our objective was therefore to quantify foliar and root uptake of DCD in pasture plants following its application under glasshouse conditions.     Gene Expression Profiling of Bronchoalveolar Lavage Cells Preceding a Clinical Diagnosis of Chronic Lung Allograft Dysfunction.

    BackgroundChronic Lung Allograft Dysfunction (CLAD) is the main limitation to long-term survival after lung transplantation. Although CLAD is usually not responsive to treatment, earlier identification may improve treatment prospects.MethodsIn a nested case control study, 1-year post transplant surveillance bronchoalveolar lavage (BAL) fluid samples were obtained from incipient CLAD (n = 9) and CLAD free (n = 8) lung transplant recipients. Incipient CLAD cases were diagnosed with CLAD within 2 years, while controls were free from CLAD for at least 4 years following bronchoscopy. Transcription profiles in the BAL cell pellets were assayed with the HG-U133 Plus 2.0 microarray (Affymetrix). Differential gene expression analysis, based on an absolute fold change (incipient CLAD vs no CLAD) >2.0 and an unadjusted p-value ≤0.05, generated a candidate list containing 55 differentially expressed probe sets (51 up-regulated, 4 down-regulated).ResultsThe cell pellets in incipient CLAD cases were skewed toward immune response pathways, dominated by genes related to recruitment, retention, activation and proliferation of cytotoxic lymphocytes (CD8+ T-cells and natural killer cells). Both hierarchical clustering and a supervised machine learning tool were able to correctly categorize most samples (82.3% and 94.1% respectively) into incipient CLAD and CLAD-free categories.ConclusionsThese findings suggest that a pathobiology, similar to AR, precedes a clinical diagnosis of CLAD. A larger prospective investigation of the BAL cell pellet transcriptome as a biomarker for CLAD risk stratification is warranted

    An Unorthodox Introduction to QCD

    These are lecture notes presented at the 2017 CTEQ Summer School at the University of Pittsburgh and the 2018 CTEQ Summer School at the University of Puerto Rico, Mayaguez. The title is a reference to hep-th/0309149 and introduces perturbative QCD and its application to jet substructure from a bottom-up perspective based on the approximation of QCD as a weakly-coupled, conformal field theory. Using this approach, a simple derivation of the Sudakov form factor with soft gluon emission modeled as a Poisson process is presented. Topics of the identification and discrimination of quark- versus gluon-initiated jets and jet grooming are also discussed.Comment: 16 pages, 18 figures.     Uninterested youth? Young people's attitudes towards party politics in Britain

    Following the outcome of the 2001 and 2005 General Elections, when the numbers of abstainers outweighed the numbers of Labour voters on both occasions, much attention has focused upon the state of British democracy and how to enthuse the electorate, especially young people. While the government is exploring ways to make the whole process of voting easier, it may be failing to tackle the real problem - that youth appear to find the business of politics uninviting and irrelevant. This paper examines data derived from a nationwide survey of over 700 young people in order to shed light on what lies at the heart of young people's apparent disengagement from formal politics in Britain - political apathy or a sense of political alienation. The findings reveal that they support the democratic process, but are sceptical of the way the British political system is organised and led, and are turned off by politicians and the political parties. However, there is no uniform youth orientation to politics, and the data indicate that views differ according to social class, educational history, and also gender. However both ethnicity and region of the country in which young people live seem to have little influence in structuring political attitudes and behaviour

    Managing and monitoring equality and diversity in UK sport: An evaluation of the sporting equals Racial Equality Standard and its impact on organizational change

    Despite greater attention to racial equality in sport in recent years, the progress of national sports organizations toward creating equality of outcomes has been limited in the United Kingdom. The collaboration of the national sports agencies, equity organizations and national sports organizations (including national governing bodies of sport) has focused on Equality Standards. The authors revisit an earlier impact study of the Racial Equality Standard in sport and supplement it with another round of interview material to assess changing strategies to manage diversity in British sport. In particular, it tracks the impact on organizational commitment to diversity through the period of the establishment of the Racial Equality Standard and its replacement by an Equality Standard that deals with other diversity issues alongside race and ethnicity. As a result, the authors question whether the new, generic Equality Standard is capable of addressing racial diversity and promoting equality of outcomes. © 2006 Sage Publications

    Speciation of phosphorus in a fertilized, reduced-till soil system: in-field treatment incubation study

Conclusive positive results of deep-banding P fertilizers compared with broadcast application and the chemistry of reduced-tillage systems remain unclear. Knowledge of the dominant solid P species present in soil following application of P fertilizers and the resulting potential P availability would help us understand and efficiently manage P in reduced-tillage systems. The objective of this research was to study the influence of placement (broadcast vs. deep-band P), fertilizer source (granular vs. liquid P), and time on the reaction products of P under field conditions. Changes in soil pH, resin-extractable P, total P, and speciation of P were determined at different distances from the point of fertilizer application at 5 wk and 6 mo after P application at a rate of 75 kg ha−1 to a soil system that was under long-term reduced tillage. Resin-extractable P was lower for broadcast treatments compared with deep-band treatments for both time periods. Resin-extractable P was greater in the liquid P-treated soils than in the granular P-treated soils. Speciation results showed that granular P fertilizers tended to form Fe–P-like forms, whereas liquid forms remained in adsorbed P-like forms in the soil 5 wk after application; moreover, speciation results showed granular P fertilizers precipitated less when deep-banded. During the 6-mo period following application, reaction products of broadcast granular, broadcast liquid, and deep-band granular fertilizers transformed to Ca-phosphate or mixtures of Ca-, Fe- and adsorbed-phosphate-like forms, whereas deep-band liquid P remained as mainly adsorbed P-like forms. Deep-banding of P would most likely provide a solution that is both agronomically and environmentally efficient for reduced-till farmers

    Mutations in pericentrin cause Seckel syndrome with defective ATR-dependent DNA damage signaling

    Large brain size is one of the defining characteristics of modern humans. Seckel syndrome (MIM 210600), a disorder of markedly reduced brain and body size, is associated with defective ATR-dependent DNA damage signaling. Only a single hypomorphic mutation of ATR has been identified in this genetically heterogeneous condition. We now report that mutations in the gene encoding pericentrin (PCNT)--resulting in the loss of pericentrin from the centrosome, where it has key functions anchoring both structural and regulatory proteins--also cause Seckel syndrome. Furthermore, we find that cells of individuals with Seckel syndrome due to mutations in PCNT (PCNT-Seckel) have defects in ATR-dependent checkpoint signaling, providing the first evidence linking a structural centrosomal protein with DNA damage signaling. These findings also suggest that other known microcephaly genes implicated in either DNA repair responses or centrosomal function may act in common developmental pathways determining human brain and body size

    Three- Versus Two-Drug Therapy for Patients With Newly Diagnosed Pulmonary Arterial Hypertension

    BACKGROUND: In pulmonary arterial hypertension (PAH), there are no data comparing initial triple oral therapy with initial double oral therapy. OBJECTIVES: TRITON (The Efficacy and Safety of Initial Triple Versus Initial Dual Oral Combination Therapy in Patients With Newly Diagnosed Pulmonary Arterial Hypertension; NCT02558231), a multicenter, double-blind, randomized phase 3b study, evaluated initial triple (macitentan, tadalafil, and selexipag) versus initial double (macitentan, tadalafil, and placebo) oral therapy in newly diagnosed, treatment-naive patients with PAH. METHODS: Efficacy was assessed until the last patient randomized completed week 26 (end of main observation period). The primary endpoint was change in pulmonary vascular resistance (PVR) at week 26. RESULTS: Patients were assigned to initial triple (n = 123) or initial double therapy (n = 124). At week 26, both treatment strategies reduced PVR compared with baseline (by 54% and 52%), with no significant difference between groups (ratio of geometric means: 0.96; 95% confidence interval: 0.86-1.07; P = 0.42). Six-minute walk distance and N-terminal pro-brain natriuretic peptide improved by week 26, with no difference between groups. Risk for disease progression (to end of main observation period) was reduced with initial triple versus initial double therapy (hazard ratio: 0.59; 95% confidence interval: 0.32-1.09). Most common adverse events with initial triple therapy included headache, diarrhea, and nausea. By the end of the main observation period, 2 patients in the initial triple and 9 in the initial double therapy groups had died. CONCLUSIONS: In patients with newly diagnosed PAH, both treatment strategies markedly reduced PVR by week 26, with no significant difference between groups (primary endpoint not met). Exploratory analyses suggested a possible signal for improved long-term outcomes with initial triple versus initial double oral therapy

    Racism, anti-racist practice and social work: articulating the teaching and learning experiences of Black social workers

    In the mid 1990s a Black practice teacher programme was established in Manchester and Merseyside with the primary aim to increase the number of Black practice teachers in social work organisations, and in turn provide a supportive and encouraging learning environment for Black student social workers whilst on placement. In the north‐west of England research has been undertaken, to establish the quality of the practice teaching and student learning taking place with Black practice teachers and students. This paper is an exploration of the ideas generated within the placement process that particularly focused on the discourse of racism and ant‐racist practice. Black students and practice teachers explain their understanding of racism and anti‐racist practice within social work. From the research, the paper will critique some of the ideas concerning anti‐racism. In particular, it will question whether anti‐racist social work practice needs to be re‐evaluated in the light of a context with new migrants, asylum seekers and refugees. It will concluded, by arguing that whilst the terms anti‐racism, Black and Minority Ethnic have resonance as a form of political strategic essentialism, it is important to develop more positive representations in the future
